Solid Dosage Formulation Principles
Binders, disintegrants, and compression science for tablet development.

Most oral medicines are still tablets or capsules. Solid dosage formulation is the craft of turning a drug substance and a short list of excipients into a unit that is manufacturable, stable, and bioavailable. Students who only memorize “binder, diluent, disintegrant, lubricant” miss the physical story: particle size, moisture, and compression force decide whether the tablet holds together on the line and still releases drug in the gut.
Functions of common excipients
Diluents (lactose, microcrystalline cellulose, dicalcium phosphate) provide bulk and compactability. Binders (povidone, hypromellose, starch paste) create granule strength in wet granulation. Disintegrants (croscarmellose, sodium starch glycolate, crospovidone) pull water in so the compact breaks. Lubricants (magnesium stearate) prevent sticking but can waterproof particles if over-blended. Glidants (colloidal silica) improve hopper flow. Coatings add taste masking, moisture protection, or delayed release — they are not decoration.
Choose the process to match the powder: direct compression when the blend flows and compresses; dry granulation (roller compaction) when the drug is moisture-sensitive; wet granulation when content uniformity or compactability is poor. Each process imprints a different granule structure and a different risk for over-lubrication or residual solvent.
Compression science in brief
- Flow: Angle of repose, Hausner ratio, and die-fill variation predict weight uniformity.
- Compactability: Tensile strength versus compression force shows the window between capping and an overly hard tablet that will not disintegrate.
- Friability and hardness: Must survive coating, packaging, and transport without failing dissolution.
- Content uniformity: Segregation of a low-dose micronized drug is a formulation problem, not only an assay problem.
- Dissolution: The in-vitro proxy for release; media, apparatus, and specification must match the product’s BCS and control strategy.
From lab tablet to robust process
Scale-up changes blending time, shear, and drying endpoint. A lubricant that was acceptable at 500 g may slow dissolution at 50 kg. Quality by design asks which material attributes (particle size, moisture) and process parameters (granulation water, mill screen, compression force) truly affect critical quality attributes (assay, dissolution, hardness). Design space is not a slogan — it is the documented range where those links still hold.
For the practicing pharmacist, formulation literacy explains real products: why a scored tablet may not be halved if the score is cosmetic, why some generics differ in excipients but not in bioequivalence, and why a patient who crushes a modified-release tablet can dump the dose. Solid dosage principles therefore connect the industrial laboratory to counseling at the counter.
